ABSTRACT

Objective@#To investigate the clinical effect of Honghua Xiaoyao granule combined with tamoxifen citrate tablets in the treatment of polycystic ovary syndrome (PCOS).@*Methods@#From June 2017 to January 2019, 92 patients with PCOS admitted to Chengde maternal and child health care hospital were selected as the study objects, and divided into control group (46 cases) and treatment group (46 cases) according to the order of admission.The patients in the control group took tamoxifen citrate tablets, 2 tablets/time, 1 time/day from the 5th day of menstrual cycle.The patients in the treatment group were treated with safflower Xiaoyao granules on the basis of tamoxifen citrate tablets, 3 bags/time, 3 times/day.After 21 days of continuous administration, 5 days of discontinuation was a course of treatment, and the two groups of patients were treated for 3 consecutive courses.The clinical effect, ovulation, blood glucose, insulin resistance index, sex hormone level and oxidative stress of the two groups were compared before and after treatment.@*Results@#The total effective rate of the treatment group was 95.65% (44/46), the ovulation rate was 97.83% (45/46), the control group was 80.43% (37/46), 84.78% (39/46), the difference between the two groups was statistically significant (χ2 value was 5.06, 4.93, P value was 0.025, 0.029). .Before and after treatment, the fasting insulin were (20.31±3.06) mU/L and (12.49±2.34) mU/L, and the homeostatic model assessment of insulin resistance were 4.84±1.30 and 2.92±0.83 in the treatment group, which were (20.14±2.55) mU/L, (16.15±2.17) mU/L, 4.86±1.08 and 3.86±0.93 in control group.There all were differences in two groups about the insulin resistance index before and after treatment, and there all were differences between two groups about the insulin resistance index after treatment (tvalue was 26.15 , 16.10, 19.68 and 12.17; all P<0.001). and there were statistically significant differences between the two indexes after treatment (t=0.77, 5.11, all P<0.001). Before and after treatment, the luteinizing hormone were (19.98±2.22) and (12.61±2.55) U/L, and the follicle stimulating hormone were (5.97±0.69) U/L and (4.52±0.79) U/L, and testosterone was (5.38±0.88) and (3.62±0.60) nmol/L, which was (20.44±2.23) U/L, (16.18±2.65) U/L, (6.09±0.59) U/L, (5.31±0.86) U/L, (5.44±0.77) nmol/L and (4.48±0.62) nmol/L in control group.The difference between the two groups before and after treatment was statistically significant (t value was 23.32, 15.29, 13.70, 8.67, 19.80 and 12.30, respectively; all P<0.001); and the difference between the groups after treatment was statistically significant (t value was 6.58, 4.61 and 6.70, respectively, all P<0.001). Before and after treatment, the malondialdehyde were (9.20±1.15) μmol/L and (5.63±0.94) μmol/L, and the superoxide dismutase were (62.99±5.37) U/L and (89.63±8.81) U/L, which were (9.45±1.08) μmol/L, (7.48±0.85) μmol/L, (61.88±5.78) U/L and (75.60±6.87) U/L in control group.There all were differences in two groups about the oxidative stress index before and after treatment, and there all were differences between two groups about the oxidative stress index after treatment (t value was 40.00, 19.84, 28.14, 15.24, 9.88 and 8.52, respectively, all P<0.001).@*Conclusion@#Honghua Xiaoyao Granules combined with Tamoxifen Citrate Tablets can effectively improved the clinical symptoms of patients with PCOS, promoting ovulation, reducing sex hormone level, improving insulin resistance and oxidative stress, which has a certain clinical application value.

ABSTRACT

Objective To observe the clinical efficacy of acupuncture plus medication and kinesiotherapy in treating cerebral stroke.Method A total of 126 patients with cerebral stroke were randomized into a treatment group of 42 cases, control group 1 of 40 cases and control group 2 of 44 cases. The treatment group was intervened by acupuncture, Chinese medication plus kinesiotherapy, control group 1 by acupuncture alone and control group 2 by kinesiotherapy alone. Before the intervention and respectively after 2-week and 4-week treatment, the three groups were observed by adopting the modified Barthel Index (MBI) and Fugl-Meyer Assessment (FMA), as well as the Activities of Daily Living (ADL).Result After the treatment, the MBI score, FMA score and ADL grading were significantly changed in the three groups (P<0.01). After 4-week treatment, the MBI score in the treatment group was significantly different from that in control group 2 (P<0.05). After 4-week treatment, the FMA score and ADL grading in the treatment group were significantly different from those in both control group 1 and 2 (P<0.05).Conclusion Acupuncture plus medication and kinesiotherapy is an effective approach in treating cerebral stroke and it can improve the ADL.

ABSTRACT

BACKGROUND:There is no effective drug for idiopathic pulmonary fibrosis (IPF), because of a lack of the animal model imitating the complete pathogenesis of human IPF. Therefore, it is critical to establish an ideal animal IPF model used for investigating the underlying pathogenesis and developing a kind of effective drug. OBJECTIVE:To establish an animal model that can mimic more characters of human IPF. METHODS:Seventy male C57BL/6 mice were randomly divided into two groups, fol owed by subjected to the intraperitoneal injection of bleomycin (35 mg/kg) on days 1, 4, 8, 11, 15, 18, 22, and 25, twice (group A) or once (group B) a week. Mice were sacrificed at 2, 4, 6, 8, and 10 weeks after the eighth injection, and the lung tissues were moved used for hematoxylin-eosin, Masson and immunohistochemical stainings. RESULTS AND CONCLUSION:There were various degrees of alveolitis and pulmonary fibrosis in the two groups at different time points after the last injection. The scores of alveolitis and pulmonary fibrosis in the group A began to gradual y increase from the 2nd week and reached the highest level at the 6th-8th weeks until the 10th week. In contrast, the scores of alveolitis and pulmonary fibrosis in the group B peaked at the 2nd week, then fluctuately decreased, and were significantly lower than those in the group A at the 6th week (P<0.05). Immunohistochemistry showed that type I col agen deposition was mainly distributed in the subpleural region, peri-vascular region and alveolar septa, which was consistent with Masson staining findings. The expression levels of transforming growth factorβ1 (TGF-β1) andα-smooth muscle actin (α-SMA) in the regions developing alveolitis and pulmonary fibrosis were significantly increased. In the group A, the expression levels of type I col agen, TGF-β1,α-SMA, and the hydroxyproline content in the lung tissues reached the peak level at 6-8 weeks. However, in the group B, al above indicators reached the highest level at the 2nd week, but gradual y decreased thereafter. At the 4th week, the expression Levels of TGF-β1 andα-SMA in the group B were significantly lower than those in the group A (P<0.05). At the 6th week, the hydroxyproline and type I col agen levels in the group B were significantly lower than those in the group A (P<0.05). In conclusion, the mouse model of pulmonary fibrosis induced by intraperitoneal injection of 35 mg/kg bleomycin twice weekly can be used to mimic the repetitive wound healing process, pathological morphology and cytokine changes of human IPF, which is prone to administration, with better stability and repeatability. ABSTRACT

This study was aimed to analyze the cold and heat nature of Chinese herbal medicine as well as its relation with the major constituent part in order to illustrate the scientific connotation of the cold and heat nature of Chinese herbal medicine from the level of material basis.There were a total of 334 kinds of Chinese herbal medicine with 280 effective ingredients recorded in the first volume of the Chinese Pharmacopoeia.The molecular descriptors of each ingredient (including one-dimensional,two-dimensional,three-dimensional,and semi-empirical calculated molecular descriptors) were calculated.The generalized rule induction (GRI) algorithm was used to mine molecular descriptors with strong correlation to the cold or heat nature of the screened Chinese herbal medicine.The results showed that molecular descriptors of molecular energy state of main components had close relationship with the nature properties of herbs.Compounds with high-energy condition were most from herbs with heat nature;while compounds with low-energy condition were most from herbs with cold nature.In addition to the energy condition of the molecule,topological molecular water partition coefficient,lipid drug property but also the main component of the polar surface area,carbon,hydrogen,oxygen atoms,the number of hydrogen bond donors and other molecular structure descriptors had certain association relationships.It was concluded that the nature of Chinese herbal medicine had certain association relationships with its main ingredients.Among them,the most important molecular descriptors of molecular energy are three-dimensional molecular descriptors.It suggested that the comprehensive effect of Chinese herbal medicine may be relevant to the energy condition of the molecule in main active components of Chinese herbal medicine and the effect on the energy metabolism of the body.

ABSTRACT

Objective To investigate the effect of hand twelve Jing-Well points bloodletting on blood-brain barrier (BBB) permeability in the early stage of focal cerebral ischemic injury in rats with permanent middle cerebral artery occlusion (pMCAO). Method Male Wister rats were randomized into sham operation, model and bloodletting groups. A model of middle cerebral artery occlusion was made by Longa's method. 2%Evans blue (EB) was injected into the caudal vein at three time points:5, 24 and 72 hrs after model making. EB exudation amount was assessed by measurement of EB absorbance in brain tissue to observe BBB permeability. Result Cerebral EB exudation amount was significantly higher in the model group of rats than in the sham operation group (P<0.01) and significantly lower in the bloodletting group than in the model group (P<0.01). Conclusion Hand twelve Jing-Well points bloodletting can reduce blood-brain barrier permeability in pMCAO rats. Its effect is highly significant at 24 and 72 hrs after ischemia.

Marine traditional Chinese medicine (MTCM) is an important part of Chinese medicine (CM),there are some differences in understanding of the current literature and the extension of the connotation of marine medicine,which leads to the definition dispute of MTCM,hindering clinical application and further development of MTCM.In this study,we explored the concept of MTCM in literature,discussed the attributes of ocean marine CM,summed up the differences between the land CM and MTCM over variety characteristics,effect of drug composition characteristics and biological activity characteristics,and discussed the connotation and extension of MTCM from three aspects of theoretical basis and the effect and source of the drug,leading to the formation of the narrow and broad concept of MTCM.The five kinds of disputes in the definition of MTCM were discriminated according to the concept and connotation,which provided a theoretical basis for the definition and the research of MTCM.Moreover,we also defined the English translation and its abbreviation as Marine Traditional Chinese Medicine (MTCM).

ABSTRACT

Twenty-nine medical records of acupuncture and moxibustion at the end of volume in Zhenjiu Dacheng are precious clinical examples. The characteristics of selecting acupoints are less and ingenious with precious matching on the basis of syndrome differentiation and examining pathogenesis detailedly, according to meridians with diseases locations recognized clearly, keeping adopting acupoints on regular meridians combined with extra points and making good use of specific points, and attaching importance to acquired essence with protecting spleen and stomach. Valuable experience is provided for physicians of later generations to treat diseases.

ABSTRACT

Marine Chinese Medicine (MCM) is one of the important part of the traditional Chinese medicine.The exploration of marine organism resources provide a good base for the development of MCM.However,the evaluation for the nature of the new source of MCM becomes one of the key problem of the clinic application of MCM.In this study,613 MCM and their related 1 091 species of marine organisms were screened.Association Rules Mining method and Phylogenetic Tree constructing method were used to find out the association relationship and the distribution regularity of the MCM with different nature in the Phylogenetic Tree.The results showed high collection of the MCM with the same nature on the tree.The MCM from the organisms in the same family might have the same nature.And the association rules for the same nature assembled at the same branch or near branch of the tree.For example,the marine plantae,Chlorophyta,Florideophyceae,and Phaeohpyceae,were related with cold nature,while the marine animalia,including Decapoda,Malacostraca,and Arthropoda,had close relationship with hot nature.Moreover,the neutral nature was related with Squamata.These results implied that there were close relationship between the nature and the affinity relationship.The MCM from the same or close families may have the same or likely nature.This study provided a new index and reference for prediction and evaluation of the nature of new MCM.

ABSTRACT

The four properties theory of traditional Chinese medicine (TCM) is an important basis for the guidance of TCM clinical medication.However,TCM modernization has produced a large number of active substances with unknown medicine properties.Therefore,it is urgent to build a new drug development system guided with TCM four properties theory.From the view of that four properties of TCM must have their material basis,and based on the known knowledge on TCM four properties,this paper proposed strategies of new drug research by cloning TCM partial components based on theTCM model of standard level property. A certain number of representative standard level property TCM drugs were selected to conduct system separation for the establishment of aTCM model of standard level property with the proportion and content of each separated component as a reference.The standard model was used as a boundary.Contents and proportions of the experimental TCM separation components deviated from thisstandard model were classified as hot or cold properties.From cloning of the partial components of TCM with hot or cold property medicine,the quality standards of partial components of experimental TCM were also studied,in order to investigate their corrective actions and mechanisms.The TCM partial components were expected to form a new TCM drug with the four properties theory of TCM as guidance.

Although the origin of Chinese medicine was well recorded inHuang-Di Nei-Jing, different opinions, however, now existed in the authenticity of discussions on five cardinal directions in theYi-Fa Fang-YiLun. From the perspective of regional systems, this paper began with the local culture of Chinese northern nomadic people, analyzed their fire using custom, moxibustion therapy, ice and snow culture and Shamanistic witchcraft. The purpose was to reveal that moxibustion therapy originated from Shamanistic witchcraft and medical practice of the northern nomadic people.

ABSTRACT

This study was aimed to analyze the research status of marine medicine in China. The research hot issues and feature rules were summarized in order to discuss development trends for the clinic application and further development of marine medicine in recent 20 years. The CNKI library was used as a source to retrieve literatures on marine medicine. The bibliometric methods and Cytoscape 3.1.1 were used to analyze. The results showed that 582 papers were included. A total of 260 papers were published during 2010 to 2014. And 52 papers were published annually. 266 papers in this field were funded. The papers were mostly published in theChinese Journal of Marine Drugs. A total of 278 research institutes were engaged in the research. And the institute published the most paper was the Ocean University of China. There were 12 researchers who had published more than 5 articles. This study focused on the keywords with highly frequency, which were marine drugs, chemical composition, marine natural products, fingerprints and antitumor. It was concluded that marine medicine had become hot issues. The core of the group has been formed. However, most researches had focused on natural products and pharmacological aspects. Traditional Chinese medicine (TCM) theory had not been taken seriously and the infrastructure was weak, which obstructed the clinical application of marine medicine. We should expand the discovery and study of knowledge on ancient herbal literature as well as clinic application of marine medicine in order to build the knowledge database of marine medicine and experience mode of clinic application.

ABSTRACT

Honeybee pupae were collected from Jilin apiaries and RNA was extracted for use as a tefnplate for amplification. Based on the complete genome sequences of black queen cell virus (BQCV) published on GenBank, we designed 10 pairs of primers to amplify genes by reverse transcription-polymerase chain reaction (RT-PCR). Using this approach, we have obtained the first complete genome sequence of a BQCV isolate in China. The genome of the isolated strain, named BQCV-JL1, is composed of 8358 nucleotides and shares between 86% and 93% homology with the complete genome sequences of the other six BQCV strains published on GenBank. ORF 1 of BQCV-JL1 is positioned between nucleot ides (nt) 546 and 4676 (4131 nt), while ORF 2 is located between nt 5750 and 8203. Between the two ORFs of BQCV-JL1 there is a short ORF, called ORF 3, between nt 4891 and 5433 (543 nt). The first functional gene ex- pression domain of the BQCV-JL1 strain is positioned between nt 546 and 5 429, encompassing both ORF 1 and ORF 3. There is an internal ribosome entry site (IRES) located before ORF 2, the last three bases of which are CCU (nt 5642-5644). These bases act as an initiation.codon facilitating the translation of ORF 2. The second functional gene expression domain of the BQCV-JL1 strain is located between nt positions 5642 and 8203. The BQCV-JL1 strain was found to share high sequence identity (93%) with the Hungary 10 genotype at the whole-genome level and analysis of the nucleotide and amino acid sequences revealed that the BQCV-JL1 strain also shows close genetic relationships with the South Korea strain, suggesting that both the BQCV-JL1 and South Korea strains may have migrated from European countries. BQCV-JL1 strain was different from the other 6 strains in dividing the nucleotides positions of QRF, which vqs because of the gene mutation.

ABSTRACT

In nature, honeybees are the most important pollinators. They play a vital role in both protecting the diversity of natural ecosystems, and maintaining the yield-improving effects of agroecosystems. But in recent years, epidemic disease in bees has caused huge losses. Black Queen Cell Virus (BQCV) is a bee pathogen that was first reported in 1955. It mainly infects bee larvae and pupae, making their bodies turn dark and black, and causing a massive decrease in the bee population. More specifically, the virus makes the exterior of the cell walls in the larvae and pupae turn black. BQCV is a seasonal epidemic, spread by means horizontal and vertical transmission, and is often unapparent. BQCV not only infects a variety of bee species, but also spiders, centipedes and other arthropods. It can also be coinfected with other honeybee viruses. In recent years, research has shown that the Nosema intestinal parasite plays an important role in BQCV transmission and bees carrying Nosema that become infected with BQCV have increased mortality. Here we summarize current research on the incidence, prevalence, geographical distribution and transmission of BQCV.

ABSTRACT

Marine medicine is an important part of Chinese medicine and a growth point of marine economy development.Exploration on the exact weight ratios and curative effect to expand marine medicine clinical application is a significant issue in marine medicine research at this stage. This paper presented a research supposition of“documents retrieval, extraction of marine medicine cases, establishment of case database, statistic analysis, data mining, herb pair Game Theory model establishment, herb pair usage under fixed dosage and matching, in order to explore a new idea of researching marine medicine combined with Game Theory. Through the literature retrieval and text extraction, 458 cases with the herb pair ofLong-Gu (LG) andMu-Li(ML) were received to establish the partnership game model. The iterative removals of strictly dominated strategies were adopted. Assuming that LG and ML used with an equal amount of 30 g was the best response, which reached the Nash equilibrium. The results suggested that dosages of LG and ML used equally at the amount of 30 g, 25 g, 24 g, 20 g, 18 g, 15 g were rationalizable strategies. Frequencies of using an equal amount of 30 g LG and ML was the most of mathematical statistics. LG and ML of 30 g were used equivalently to treat symptoms such as insomnia, inappetence, chills, tinnitus, physically and mentally fatigued, spontaneous sweating and night sweating, and so on with the effect of tranquilizing and constricting. The results can be validated and complemented among association rules analysis, mathematical statistics and game model. It implied that it was feasible to apply mathematical statistics and data mining in combination with Game Theory to study the marine medicine herb pair.

ABSTRACT

The number of hypertension patients in China has increased year by year. Traditional Chinese medicine (TCM) has obvious advantages in the treatment of hypertension. Document resources of TCM treatment on hypertension play an important role as references and guidance. Agile construction thinking, architecture based on MVC, and data demand driven will make the building of information service system with efficiency and high quality. The content provided by this information service system will promote the scientific research and clinical practice of hypertension.

ABSTRACT

Experience mode of marine medicine (EMMM) is the summary of experiences on marine medicine con-cluded by ancient doctors for thousands of years. The mining of EMMM is very important for the clinic application and development of marine medicine. However, these experiences are very difficult to discover for they are all scat-tered in the large number of ancient documents. We proposed that the EMMM can be built by the procedure of in-formation of ancient documents, text-mining of ancient documents, and associate network of informatics unit of ma-rine medicine by non-interactive literature-based knowledge discovery methods, and expert evaluation at the end, so as to provide database for the clinic application and further development of marine medicine.

ABSTRACT

Objective To comparative the expression of suppressors of cytokine signaling 3 (SOCS3) in placental tissues between premature delivery and term delivery,and to evaluate the significance of SOCS3 in premature-delivered placental tissues.Meth ods Retrospective analysis was made on 30 patients who had spontaneous premature delivery and 30 pregnant women with normal delivery from February 2013 to October 2013 in our hospital.Reverse transcription and polymerase chain reaction (RT-PCR) was used to detect the mRNA expressions of SOCS3,interferon-α(IFN-α),and interleukin-10 (IL-10).Western-blot was used to detect protein expression of SOCS3,and to analyze the balance of Th1/Th2.Results PCR detection showed that SOCS3-mRNA in the control group was significantly lower than that in the experimental group (P < 0.01).Western-blot test showed that SOCS3 protein expression in the control group was significantly lower than that in the experimental group (P < 0.01).The expression of SOCS3 gene was consistent with its protein expression.PCR detected the significant increase of IFN-α with the secretion of Th1 cytokine,significant decrease of IL-10 with secretion of Th2 cytokine (P < 0.05),and the balance deviation of Th1/Th2 toward Th1.Conclusions SOCS3 is associated with premature delivery,which mechanism might be involved in regulation of the immune balance through janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathways.

ABSTRACT

Marine medicine is an important part of Chinese medicine and a growth point of the development of marine economy. Current researches of marine medicine had ignored accumulated experience in clinical medicine for thousands of years of development of marine medicine and the guidance of traditional Chinese medicine (TCM) theory. In this article, we proposed a research mode of Mining experience-Clinical validation-Targets screening-Component screening-Component compatibility-Mechanism study under the guidance of the hypothesis of Nature-Effect-Material Trialism to study marine medicine. Kun Hai Jiang Xin (KHJX) Decoction is a marine formula drawn from the experience of ancient physicians by data mining techniques. Clinical study found that KHJX Decoction can significantly improve the overall clinical efficacy of chronic bronchitis with good clinical safety. Under the guidance of TCM herbal theory, object presented here is based on the work of KHJX Decoctionmining experience - clinical validation. The molecular docking method and receptor-ligand binding assay were applied to screen effective components from herbs of KHJX Decoction and the compatibility plan of effective components under the guidance of the principle of cold and heat effects compatibility. This study is of great significance to clarify the material basis and molecular mechanism of KHJX Decoction in the treatment of chronic bronchitis. It provides a new research mode for marine drug research.

ABSTRACT

Aim: To study the synthesis and anti-inflammatory activity of p-(sulfamyl) benzylidene-linked hetero-cyclic ketone derivatives. Methods: A series of p-(sulfamyl) benzylidene-linked heterocyclic ketone derivatives were synthesized. Anti-inflammatory activity was evaluated against xylene-induced ear oedema in mice and against carrageenan-induced paw oedema in rats. Gastrointestinal side effects in the rats were also examined after continu-ous introgastric administration of these compounds once daily for 7 days. Results: Twelve compounds( LHZ-101-LHZ-112) were synthesized and their structures were confirmed by IR, ~1H NMR, MS and elemental analysis. LHZ-105, LHZ-106 and LHZ-111 exhibited marked anti-inflammatory activity in xylene-induced mice ear swelling model. LHZ-106 and LHZ-111 showed significant anti-inflammatory activity in carrageenan-induced rat paw ede-ma model. LHZ-105, LHZ-106 and LHZ-111 had less gastrointestinal side effects than diclofenac sodium and CI-1004. Conclusion: These results suggest that some of these compounds have the potential for anti-inflammatory activity with few gastrointestinal side effects.
